The Population Tester Kit contains 9 highly polymorphic microsatellite markers that are designed for laboratories interested in strain identification and gene flow questions, or those groups that are experimenting with the possible use of molecular markers on a limited basis. For those wishing to employ these chicken primers to yield polymorphic products in other avian species, your likelihood of success will depend greatly on how divergent that species is from chicken and on PCR conditions. We and others have had mixed success in applying chicken microsatellite primers in the turkey, but relatively little success in limited experiments with other species. (While primer sets may amplify a band from other avian DNAs, such PCR products don't necessarily derive from a locus homologous to the corresponding chicken locus.)
These primers were synthesized and are provided with support from the NAGRP, (USDA-CSREES, NRSP-8), a generous gift from Hy-Line International and in-kind support for Poultry Genome Coordination from the USDA-ARS Avian Disease and Oncology Laboratory (ADOL) and Michigan State University. (Please note: The general use of microsatellite primers is covered by one or more patents to the Marshfield Clinic. NAGRP obtains its primers from a licensed supplier (ResGen/Invitrogen), and Marshfield is paid a fee for the right to distribute this kit only for non-profit purposes or to companies that provide evidence that they are separately licensed by Marshfield for microsatellite applications. In addition, MCW markers up to and including MCW193 are the subject of patent applications by Euribrid B.V. Inquiries about the accepted use of these primers should be directed to Euribrid as noted in Poultry Science 75:746-754, 1996. The kit is offered with the understanding that appropriate use of potentially patented material is the responsibility of the user and not of the Poultry Coordinators.)
